For Modbus Serial communication between M241/M251/M262 and Altivar 320, does the Polarization Resistor setting need to be turned on?

Yes, polarization must be turned on in the PLC as per p27 of the ATV320 Modbus Manual:

To apply the setting, in EcoStruxure Machine Expert (or the previous version SoMachine):
- In the Devices Tree of your EcoStruxure Machine Expert / SoMachine project, expand the tree for your PLC.
- Doubleclick on the Serial_Line_1 (or Serial_Line_2)node
- Under "Physical Medium", choose "Yes" in the dropdown list for Polarization Resistor
