Does a 2 Pole Main breaker need to be used to change a 400 amp I-Line 3 phase panel, to a single phase Panel?

No. A 3 pole Main breaker can still be used for single phase, simply by removing the B phase lug from the main breaker, and wiring to A and C phases only.
NOTE: All branch breakers must be single pole or two pole and can only be connected to A and C phases.
